Kisspeptin and the HPG Axis: Why the Dosing Schedule Looks So Different

Every other protocol on this site doses once or twice a day at most. Kisspeptin-10 is dosed several times a day — and that's a direct consequence of where it sits in the hormone cascade.

The hypothalamic-pituitary-gonadal (HPG) axis is the cascade that governs reproductive hormone production, and it starts at the very top: the hypothalamus releasing GnRH (gonadotropin-releasing hormone) in pulses, which then triggers the pituitary to release LH and FSH, which in turn drive downstream sex-hormone production. Kisspeptin sits one step upstream of GnRH itself — it's the signal that tells GnRH-releasing neurons to fire.

That upstream position is exactly why kisspeptin's dosing schedule looks so different from everything else on this site. GnRH itself is only released in short pulses, and the axis is designed around pulsatile signaling, not a sustained baseline level — continuous, non-pulsed GnRH exposure actually desensitizes the pituitary and shuts the axis down rather than stimulating it, which is the mechanism paradoxically exploited by some GnRH-agonist suppression therapies. Kisspeptin-10 also has a very short half-life, on the order of minutes, so a single dose produces a brief signal rather than a sustained one.

Put those two facts together — an axis that responds to pulses rather than sustained signal, and a peptide that's naturally cleared in minutes — and dosing several small amounts throughout the day rather than one larger daily dose isn't just a convention, it's an attempt to mimic the pulsatile pattern the axis is actually built to respond to.

This is also why baseline hormone bloodwork matters more here than for most other compounds discussed on this site: kisspeptin is acting on the control point for the entire reproductive hormone cascade, not a downstream or peripheral target, so understanding where that axis already sits before intervening on it is a meaningfully different risk calculation than, say, a topical skin peptide.
